FOXO4-DRI protocol

FOXO4-DRI is a lab-designed peptide that makes aged ('senescent') cells self-destruct. It has been tested in cells and mice. There are no human data.

The protocol at a glance

No human dose exists. Mouse papers gave 5 mg per kg into the belly cavity every other day for three doses.

Community reports

Community reports commonly describe either an escalating 250-500 mcg daily plan or 3 mg every other day for 3 doses.

What people say they do, collected from community discussions. Not tested in a trial; shown so you can compare it with the evidence above.

Mixing

No mixing table here: this one is used as a capsule, nasal spray or skin product, or has no human amount to calculate from. The general mixing calculator works for any vial.

What the trials found

There are no human results, so these are the evidence facts, all from mice and cells:

5 mg/kgMouse dose, every other day, 3 times
30 daysTime after treatment when aged mice had higher testosterone (2020 paper)
3Mouse and cell papers checked

Baar MP et al. Cell 2017 (PMID 28340339): in aged and fast-aging mice, restored fitness, fur density and kidney function. Aging 2020 (PMID 31959736) and J Cell Mol Med 2022 (PMID 35510614) are later mouse studies.

Side effects

  • Side effects in people are unknown.
  • The 2017 paper says it was well tolerated in mice at the doses used. Because the peptide is built to kill cells, its effects on healthy tissue in people have not been tested.

How it works

FOXO4-p53 link

Aged cells use FOXO4 to hold p53 inside the nucleus and stay alive. FOXO4-DRI blocks that link, so p53 leaves the nucleus and those cells die by apoptosis.
