Humanin protocol

Humanin is a small protective peptide made from mitochondrial DNA. It has been studied in cells and in mice and rats. No dosing trial in people was found.

The protocol at a glance

No human dose exists. Animal work used humanin analogs such as HNG (S14G-humanin) and HNGF6A, which are not the same molecule as natural humanin.

ItemFinding
Human doseNot established
Studied inMice and rats (belly injection, into a vein, into the brain) and cell cultures
Human dataBlood levels measured in observational studies; no dosing study found

Mouse and rat amounts are not shown because they do not convert to a human dose. Animal studies mostly used the analogs HNG or HNGF6A.

Community reports

Community reports commonly describe 25 mcg to 1 mg, with some confusion between humanin and the HNG analog.

What people say they do, collected from community discussions. Not tested in a trial; shown so you can compare it with the evidence above.

Mixing

No mixing table here: this one is used as a capsule, nasal spray or skin product, or has no human amount to calculate from. The general mixing calculator works for any vial.

What the trials found

There are no human treatment results, so these are the evidence facts:

3 monthsHNG injections in Alzheimer's-model mice: better memory and fewer amyloid plaques
83Dialysis patients in a 2026 study: blood humanin measured, and both low and high levels were linked to more heart events
49Older adults in a 2025 study: higher blood humanin with better Mediterranean-diet adherence

Frontiers in Nutrition 2025 (PMID 41883858); Cardiorenal Medicine 2026 (PMID 41849628); Pharmacol Biochem Behav 2012 (PMID 21993310). These human studies only measured natural blood levels.

Side effects

  • Side effects in people are unknown because humanin has not been given as a drug in a human trial that I found.

How it works

Cell-protection signals

Humanin binds a receptor made of three proteins and turns on signals (JAK2/STAT3 and SH3BP5) that block nerve cell death in lab models of Alzheimer's.

Insulin and fat handling (animals)

Analogs improved insulin response and, in mice on a high-fat diet, lowered weight gain, belly fat and liver fat.
