The protocol at a glance
In the phase 3 trial, people injected survodutide weekly, with the amount raised slowly to a top of 3.6 mg or 6.0 mg, for 76 weeks.
| Study | Weekly amount |
|---|---|
| Phase 3 obesity, 76 weeks | Raised up to 3.6 mg or 6.0 mg (step-up plan not in the sources I checked) |
| Phase 2 obesity, 46 weeks | 0.6, 2.4, 3.6 or 4.8 mg (20 weeks raising, 26 weeks steady) |
| Phase 2 liver disease (MASH), 48 weeks | 2.4, 4.8 or 6.0 mg (24 weeks raising, 24 weeks steady) |

Community notes tend to copy trial numbers and start low. This is community talk, not a label.

What the trials found
SYNCHRONIZE-1: 725 adults with obesity, no diabetes, 76 weeks. Average body weight change (treatment-regimen analysis, which includes people who stopped early):
NEJM 2026 (SYNCHRONIZE-1, NCT06066515). The company's earlier headline of up to 16.6% used a different analysis (efficacy estimand, assumes the drug was taken as planned).
Side effects
- Stomach and gut effects were the most common: 81% on 3.6 mg and 90% on 6.0 mg, versus 48% on placebo. They were usually mild to moderate. No deaths were reported.
- In the liver trial, nausea was 66% vs 23% on placebo, diarrhea 49% vs 23%, and vomiting 41% vs 4%.
How it works
GLP-1 receptor
Switches on the GLP-1 receptor, which lowers appetite.
Glucagon receptor
Also switches on the glucagon receptor. The liver trial studied it for fatty liver disease (MASH).
